Immunogen: Recombinant protein corresponding to Arg23-Leu502 from mouse Sulfamidase, expressed in Sf 21 cells.
Also known as N-sulfoglucosamine sulfohydrolase and heparan N-sulfatase, Sulfamidase/SGSH is an important member of the sulfatase family involved in the degradation of heparan sulfate (1). The SGSH deficiency results in mucopolysaccharidosis type IIIA (MPS IIIA, Sanfilippo A syndrome), an autosomal recessive lysosomal storage disease characterized by neurological dysfunction but relatively mild somatic manifestations (2). The deduced amino acid sequence of mouse SGSH consists of a signal peptide (residues 1-22) and a mature chain (residues 23-502) (1). rmSGSH corresponds to the mature chain and has sulfatase activity. Applications: Suitable for use in Western Blot, Immunoprecipitation and ELISA. Other applications not tested. Recommended Dilution: Western Blot: 0.1-0.2ug/ml Detects mouse SGSH. The detection limit is ~5ng/lane under non-reducing and reducing conditions. Immunoprecipitation: 25ug/ml Immunoprecipitates rmSGSH from conditioned media of transfected NS0 cells. Direct ELISA: 0.5-1.0ug/ml Detects mouse SGSH. The detection limit for rmSGSH is ~0.3ng/well. Optimal dilution determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
