A DNA sequence encoding the signal peptide from human CD33 joined with the extracellular domain of human IGSF4D (Ala 21-Asp 335) was fused with a 6X histidine tag at the C-terminus. Molecular Mass Based on N-terminal amino acid sequencing, the recombinant human IGSF4D protein starts at Ala 21 and has a predicted molecular mass of approximately 35.5kD. As a result of glycosylation, the recombinant monomer migrates as a an approximately 36-55kD protein on a SDS-PAGE gel.Immobilized rhIGSF4D (at a 3il droplet containing 50 ng) on a nitrocellulose coated microplate is sufficient to significantly enhance neurite outgrowth. Human IGSF4D IGSF4D is an immunoglobulin superfamily member that is also designated as synaptic cell adhesion molecule 2 (SynCAM 2) and nectin-like protein 3 (Necl-3). The four IGSF-4 proteins, designated A, B, C and D, are type I transmembrane glycoproteins expressed mainly in epithelial cells that contain one V-type Ig-like and two C2-type Ig-like domains in their extracellular regions. These domains are characteristically responsible for Ca-independent homophilic and heterophilic interactions in the brain, lung, kidney, bladder, prostate and testis. Amino acids found to be critical in other IGSF-4 proteins for adhesion and intracellular binding of MAGUK guanylate kinase subfamily proteins are conserved in IGSF4D. The expression and function of IGSF4D has not been well characterized. However, like other family members, IGSF4D has a positive effect on in vitro outgrowth of cortical neurons. Hybridoma: NSO myeloma cells with spleen cells from Balb/c mice. Endotoxin: < 1.0 EU per 1ug of protein as determined by the LAL method. Storage and Stability: Lyophilized powder may be stored at -20C. Reconstitute to nominal volume by adding sterile PBS. For long term storagea carrier protein (e.g. 0.1% HSA or BSA) should be added to the vial. Aliquot and store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
