Iodide (I-) is an essential constituent of thyroid hormones. Thyroid epithelial cells for the biosynthesis of thyroid hormones actively transport it. At the cellular level, Iodide transport is mediated by intrinsic membrane Na+/I+ symporter (NIS/TIT). Rat TIT/SLC5A (618aa, ~66kD, mouse 618aa, human 643aa) is predicted to contain 12 transmembrane domains. The N-terminus has a short extracellular domain, whereas C-terminus is predicted to be cytoplasmic. TIT is primarily expressed in thyroid but also detected in mammary gland and ovary. Its expression is reduced in tumors. Defects in TIT gene are associated with iodide transport defect (ITD), which is characterized by an inability of the thyroid to maintain concentration difference of readily exchangeable iodine between plasma and the thyroid gland. It is a cause of congenital hypothyroidism. Applications: Suitable for use in ELISA, Immunofluorescence and Immunohistochemistry. Other potential application, though not tested is Western Blot. Other applications not tested. Recommended Dilutions: ELISA: 1:100,000 using 50-100ng control peptide/well Immunohistochemistry (Formalin-fixed): 2-20ug/ml Optimal dilutions to be determined by the researcher. Control Peptide: T5610-02: Thyroid Iodide Transporter, Rat (TIT) (Control Peptide) Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
