A 16aa peptide sequence (designated TIT11-P, control peptide), near the C-terminal, cytoplasmic domain of rat TIT. The 16-aa rat peptide sequence is unique to TIT and no significant sequence is seen with glucose transporter or other Na+-dependent transporters. Iodide (I-) is an essential constituent of thyroid hormones. Thyroid epithelial cells for the biosynthesis of thyroid hormones actively transport it. At the cellular level, Iodide transport is mediated by intrinsic membrane Na+/I+ symporter (NIS). Rat TIT (618 aa, ~66kD) is predicted to contain 12 transmembrane domains. Both N and C-termini are predicted to be cytoplasmic. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
