Activation of apoptosis by CD253 occurs through binding to the death-signaling receptors DR4 (TRAILR1) and DR5 (TRAILR2), leading to formation of the death-inducing signaling complex (DISC) and the triggering of a proteolytic caspase cascade. There are however decoy receptors known as DcR1 (TRAILR3) and DcR2 (TRAILR4) which competitively bind to CD253 and are unable to initiate apoptosis. CD253 also plays a role in immune surveillance, acting as a mediator of monocyte, NK cell, dendritic cell and T cell anti-tumor and anti-viral cytotoxicity. Applications: Suitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (paraffin): 0.25ug/ml ELISA: 0.5ug/ml Western Blot: 0.1-0.2ug/ml Requires antigen retrieval using heat treatment for 20 minutes prior to staining of paraffin sections. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
