TCF7 (Transcription factor 7, also T cell factor 1/TCF1) is a 25-50kD member of the lymphoid enhancer binding factor family of proteins with 16 isoforms. It is expressed in thymocytes and mature T cells, and serves multiple purposes. In resting cells, TCF family members are transcriptional repressors, and are 25-32kD in size. Following activation, large TCF7 isoforms predominate (42-50kD), and serve a transcriptional activator function. Human TCF7 is 384aa in length. This is likely an activating isoform that contains a beta-catenin binding domain (aa1-59), a DNA-binding HMG-box (aa269-337), and an NLS (aa344-348). The use of an alternate start site at Met116 seems to characterize repressor isoforms. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.2ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
