Transforming growth factor-beta (TGFb) family of proteins has potent hematopoietic regulatory properties ranging from effects on the growth and differentiation of primitive stem cells to the differentiated functions of mature cells. Although most reports have described the inhibitory activities of TGF-beta on hematopoiesis, recent evidence supports the concept that TGF-beta can have both inhibitory and stimulatory actions on these systems. These differences depend on the differentiation state of the target cell and the other cytokines interacting with the cell. Furthermore, TGF-beta has direct bidirectional effects on cell surface expression of many cytokine receptors suggesting that it is part of the mechanism of action of TGF-beta. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry and Neutralization. Other applications have not been tested. Recommended Dilutions: Western Blot: 1ug/ml using Recombinant Human LAP TGF-beta 1. Immunohistochemistry: 5-15ug/ml using immersion fixed paraffin-embedded sections of human prostate cancer tissue. Immunocytochemistry: 5-15ug/ml using immersion fixed human peripheral blood mononuclear cells (PBMCs). Neutralization: Measured by its ability to neutralize LAP TGF-beta 1 inhibition of TGF-beta 1 growth inhibition in the HT-2 mouse T cell line. Tsang, M. et al. (1995) Cytokine 7:389. The Neutralization Dose (ND50) is typically 15-30ug/mL in the presence of 125ng/mL Recombinant Human LAP TGF- beta 1 and 0.25ng/mL TGF- beta 1. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
