Immunogen: Recombinant protein corresponding to Ala22-Pro320 from human TRAP 5 expressed in NSO cells (NP_001602).
Encoded by the ACP5 gene, Tartrate Resistant Acid Phosphatase (TRACP or TRAP) is also known as Purple Acid Phosphatase (PAP) or Acid Phosphatase 5 (ACP5). The deuced amino acid (aa) sequence of human ACP5 predicts a signal peptide (aa1-21) and a mature chain (aa22-325). Recombinant human ACP5 consists of aa22-320, without the last 5 residues, RRARP. TRAP 5 is expressed at high levels by osteoclasts, macrophages and dendritic cells. Two forms, 5a and 5b, circulating in human blood, are derived from different cell types and have different functions. Derived from macrophages and dendritic cells, 5a is a marker of inflammatory conditions. Derived from osteoclasts, 5b is a marker of bone resorption. Compared to 5a, 5b does not contain sialic acid residues, has a higher specific activity and pH optimum, and may be processed into a disulfide-linked dimer. Applications: Suitable for use in Western Blot, Immunoprecipitation and Direct ELISA. Other applications not tested. Recommended Dilutions: Western Blot: 0.1-0.2ug/ml. The detection limit for recombinant human TRAP 5 is ~25ng/lane under non-reducing and reducing conditions. Immunoprecipitation: Immunoprecipitates recombinant human TRAP 5 from conditioned media of transfected NSO cells. Direct ELISA: 0.5-1ug/ml. The detection limit for recombinant human TRAP 5 is ~0.3ng/well. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
