Minor disruption of the GI surface layer of the cells occurs frequently, such that maintaining epithelial integrity is of crucial importance. Normal epithelial repair requires restitution and regeneration. During restitution, within minutes after injury, epithelial cells spread and migrate across the basement membrane to re-establish surface cell continuity. Trefoil factor family (TFF) proteins play an important role in restitution by influencing the migration of cells without promotion of proliferation or tumorigenesis. Regeneration occurs later on and involves proliferation and differentiation of epithelial cells and restoration of specialized elements. Three mouse TFFs have been identified to date: TFF1 (formerly pS2) and TFF3 (formerly hITF), with one trefoil domain each, and TFF2 (formerly hSP), with two trefoil domains. TFF2: rat, human and mouse: 129aa each - 14.1kD, mouse chromosome: 17A3.3. In normal gastric mucosa, TFF2 expression is observed in mucous glands of body and antrum. In duodenum, TFF2 expression is present in Brunners glands acini and distal ducts. Some focal expression is also observed in duct epithelium of pancreas and in gall bladder epithelium. Applications: Suitable for use in Western Blot, ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1-10ug/ml (Chemiluminescence) ELISA: 1:100,000, 50-100ng of control peptide/well Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
