Human troponin complex
Troponin, working in conjunction with tropomyosin, functions as a molecular switch, regulating muscle contraction in response to changes in the intracellular Ca2+ concentration. Troponin consists of three subunits, the Ca2+-binding subunit troponin C (TnC), the tropomyosin-binding subunit troponin T (TnT), and the inhibitory subunit troponin I (TnI). Troponin I binds to actin in thin myofilaments to hold the actin-tropomyosin complex in place Because of which myosin can not bind the actin in relaxed muscle. When Calcium binds to the Troponin C it causes certain confirmational changes which leads to dislocation of troponin I and finally tropomyosin leaves the the binding site for myosin on actin leading to contraction of muscle. The letter I is given due to its inhibitory character. Applications: Suitable for use in Western Blot and Sandwich ELISA. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Hybridoma: Sp2/0 myeloma cells with spleen cells of Balb/c mice. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
