MaxLight(TM)650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor(TM)647, DyLight(TM)649, Cy5(TM) and offers better labeling efficiency, brighter imaging and increased immunodetection. Human cardiac-specific troponin I (TnI) is an integral inhibitory protein of cardiac muscle which exists as a complex with troponin C (TnC) and troponin T (TnT). The TnT subunit of troponin binds to tropomyosin to form a troponin-tropomyosin complex, anchored in place by the binding of TnI to actin, within muscle thin filaments. Structural change resulting from the binding of calcium to specific sites on the regulatory TnC subunit, releases the inhibitory region of TnI from actin, enabling the attachment of the molecular motor protein myosin, allowing for muscle contraction and hence movement. The measurement of blood TnI and TnT levels is an important diagnostic indicator of heart muscle damage, and can be used to differentiate between angina and myocardic infarction in patients with chest pain. Troponin I has also been shown to inhibit angiogenesis in vivo and in vitro. Applications: Suitable for use in FLISA, Immunohistochemistry, Western Blot and Immunofluorescence. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight(TM)650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
