SK, also known as Tsukushi and Leucine-rich repeat-containing protein 54, is a member of the small leucine-rich proteoglycan (SLRP) family of extracellular matrix proteins, which are characterized by leucine-rich repeats (LRR) flanked by conserved cysteines. Mouse TSK is a secreted glycoprotein that has a predicted molecular weight of 36.5kD and contains 9 LLR. Proteoglycan modification of TSK has not been shown. Estrogen treatment in breast cancer cell lines up-regulates TSK, and so TSK is proposed to be involved in hormonally regulated extracellular matrix remodeling. Mouse TSK shares 96%, 85%, 51%, and 49% sequence identity with rat, human, chicken, and Xenopus TSK, respectively. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Direct ELISA: 0.5-1ug/ml Western Blot: 1-2ug/ml Optimal dilutions to be determined by the researcher. Hybridoma: Myeloma cells with B cells from Balb/c mice. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
