Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. Applications: Suitable for use in ELISA, Western Blot and Neutralization. Other applications not tested. Recommended Dilution: Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of mTNF-alpha (0.5ng/ml), a concentration of 0.010-0.015ug/ml of this antibody is required. ELISA: To detect mTNF-alpha by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2-0.4ng/well of recombinant mTNF-alpha. Western Blot: To detect mTNF-alpha by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant mTNFalpha is 1.5-3.0ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
