Tumor Necrosis Factor-alpha (TNF-alpha) is a pleiotrophic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF superfamily of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, autoimmune diseases, rheumatoid arthritis, inflammation and diabetes. Applications: Suitable for use in ELISA, Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistology (Frozen): 1:50. 6um tissue sections fixed in acetone for 10 min (4). Flow Cytometry: 1:50. Cells were fixed in PBA containing 0.2% formaldyde (1). Western Blot: 1:50 detects a band at ~17kD. A reduced sample treatment and 15% SDS-PAGE were used. (4) Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
