Tumor Necrosis Factor alpha (TNF-alpha) is a 17.4kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. Application: Suitable for use in ELISA and Western Blot. Other applications have not been tested. Recommended Dilution: ELISA: 0.2-0.4ug/ml (using 100ul/well) Used in conjunction T9160-27 as a capture antibody, allows the detection of at least 0.2ng/well of recombinant hTNF-a. Western Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hTNF-a is 1.5-3.0ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS, 0.1% BSA. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
