Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. Applications: Suitable for use in ELISA, Western Blot and Neutralization. Other applications have not been tested. Recommended Dilution: ELISA: 0.5ug/ml (100ul/well antibody solution). Detection of 0.2-0.4ng mTNFa/well. Western Blot: 0.10.2ug/ml. Detection limit is 1.5-3ng mTNFa/lane, under either reducing or non-reducing conditions. Neutralization: To yield one half maximal inhibition (ND50) of the biological activity of mTNF-alpha (0.25ng/ml), a concentration of 0.05-0.08ug/ml of this antibody is required. Optimal dilutions to be determined by the researcher. Reconstitution: Reconstitute in sterile water to a concentration of 0.2mg/ml. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Country of Origin: USA
