Recombinant protein corresponding to human tumor necrosis factor receptor type 1.
The immunoregulatory cytokine tumor necrosis factor alpha (TNFa) recognizes two cell surface receptors. TNF-R1 (also known as CD120a) has an apparent molecular mass of 55/60kD and is constitutively expressed on all nucleated cells, whereas TNF R2 (also known as CD120b) with an apparent molecular mass of 75/80kD, is restricted to hematopoietic cells. TNF-R2 directs TNFa mediated activation and proliferation of T lymphocytes. An autocrine role for TNF and TNF-R2 is implicated in hematopoietic malignancies, and the TNF-R2 locus is a translocation target in several leukemias, suggesting a role for TNF-R2 in the development of certain leukemias. Applications: Suitable for use in ELISA, Radioimmunoassay and Western Blot. Other applications have not been tested. Recommended Dilutions: Western Blot: detects high levels of TNFR1 in recombinant material under reducing conditions, but does not detect TNFR1 in normal cells. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
