MaxLight(TM)550 is a new Yellow-Green photostable dye conjugate comparable to Alexa Fluor(TM)546, 555, DyLight(TM)549 , Cy3(TM), TRITC and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (550nm), Emission (575nm), Extinction Coefficient 150,000. TNF-related weak inducer of apoptosis (TWEAK) is a member of the tumor necrosis factor superfamily (TNFSF) of structurally related cytokines. Like most other members of this family, TWEAK is a cell surface-associated type II transmembrane protein although a smaller, biologically active form can also be generated by cleavage near the cell membrane. TWEAK has multiple biological activities, including stimulation of cell growth and angiogenesis, induction of inflammatory cytokines, in addition to stimulation of apoptosis. The TWEAK signal transduction pathway has not been well established but it appears to signal via TweakR/Fn14 in a manner similar to that described for other TNFSF members that bind receptors lacking death domains. Applications: Suitable for use in FLISA and Western Blot. Other applications not tested. Recommended Dilutions: Direct FLISA: 0.5ug/ml using 100ul/well antibody solution detects 0.2-0.4ng/well of recombinant human TWEAK. Western Blot: 0.1-0.2ug/ml detects 1.5-3.0ng/lane, under either reducing or or non-reducing conditions. Optimal dilutions to be determined by researcher. Storage and Stability: May be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Note: Applications are based on unconjugated antibody.
