Tyrosine hydroxylase (TH) is the rate-limiting enzyme in the synthesis of the catecholamines dopamine and norepinephrine. TH antibodies can therefore be used as markers for dopaminergic and noradrenergic neurons in a variety of applications including depression, schizophrenia, Parkinsons disease and drug abuse (Kish et al., 2001, Zhu et al., 2000, Zhu et al., 1999). TH antibodies can also be used to explore basic mechanisms of dopamine and norepinephrine signaling (Witkovsky et al., 2000, Salvatore et al., 2001, Dunkley et al., 2004). Applications: Suitable for use in Western Blot, Immunohistochemistry, Immunofluorescence and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000, Detects a band of ~60kD in rat caudate cell lysates. Immunohistochemistry: 1:1000 Immunocytochemistry (IF): 1:500-1:1000, fixed with 4% PFA and permeabilized with 0.1-0.3 % Triton X. Optimal dilution determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
