Immunogen: Synthetic peptide corresponding to the N-terminal sequence of human Tyrosine Hydroxylase.
Tyrosine hydroxylase catalyzes the rate limiting step in the synthesis of dopamine and other catecholamines. The enzymes activity is regulated at the levels of transcription, translation and posttranslational modification (1,2). Tyrosine hydroxylase exists in several different isoforms ranging from 50kD to about 62kD in human. The N-terminal regulatory domain contains three phospho-serine sites: serine 19, serine 31 and serine 40 (4,5). Phosphorylation at serine 40 by PKA positively regulates the enzyme catalytic activity of the enzyme (4-6). Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in TBS, 5% BSA, 0.1% Tween-20 at 4C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
