Product Description

U73122 inhibits agonist induced phospholipase C (PLC) activation in human platelets and neutrophils. This compound also inhibits agonist-induced down-regulation of muscarinic receptors in SK-N-SH neuroblastoma cells. U73122 is a useful tool for investigating receptor-mediated PI turnover in signal transduction and is a potent inhibitor of human neutrophil adhesion to biological surfaces, as well as adhesion-dependent granule exocytosis and oxidative burst. Biological Activity: IC50 = 1.0-2.1uM (agonist induced PLC activation in human platelets and neutrophils) Solubility: Soluble in chloroform, soluble in methylene chloride, soluble in DMSO at a concentration of 2mg/ml, and soluble in ethanol at a concentration of 1mg/ml. Suggested Reconstitution: Because U73122 is only slightly soluble in aqueous media, it is best delivered to cells either when dissolved in organic solvents or when complexed with serum proteins. The following methods of reconstitution and delivery to cells are recommended. Method 1: 1. U73122 may be dissolved directly in ethanol or DMSO immediately prior to use. Unused portions may be stored at -20C for up to two months. Method 2: 1. Weigh out the quantity of U73122 required for the particular experimental application and dissolve the compound in chloroform or methylene chloride using appropriate solvent-resistant tubes. 2. Apportion the dissolved U73122 into working aliquots and evaporate the solvent under a nitrogen stream. A thin dry film will be visible. 3. The film may be dissolved in DMSO (solubility: 5 mM) or 100% ethanol (solubility: 2 mM). Alternatively, the film may be dissolved with aqueous medium containing serum or serum albumin at a concentration of 1-10mg/ml. Sterility: This product is not sterile. Storage: Store, as supplied, at room temperature. Upon solubilization, apportion into working aliquots and store at -20C. Avoid repeated freeze/thaw cycles. Solutions are stable at -20C for up to two months. Discard solutions that have turned a pink color, as this indicates a loss in inhibitory activity.
