Human Ubquitin Conjugating Enzyme 7 (UbcH7) is a class I enzyme which functions in the stress response and the control of transcription factors. The enzyme is ubiquitously expressed with high levels of expression seen in adult muscle. UbcH7 mediates the selective degradation of short-lived and abnormal proteins and is highly homologous to UbcH5. It has been demonstrated to participate in the ubiquitinylation of p53, c-Fos and NF- B. UbcH7 is one of two E2s (UbcH5 being the other) with which HECT domain proteins interact with UbcH7 being able to efficiently substitute for UbcH5 in E6-AP-dependent ubiquitinylation. Recombinant Human Ubiquitin Conjugating Enzyme 7 produced in E.coli is a 18.9kD protein containing 162 amino acids The rhUbcH7 protein contains 6xHis tag and is purified by proprietary chromatographic techniques. Recombinant protein corresponding to aa2-end of human UbcH7 with N-terminal His-tag expressed in E. coli (NM_003347). Specific Activity: 50ul reaction mix (4mM ATP, 5mM MgCl2, 1mM DTT, UBE1 (0.5ug), UbcH7 or other E2, Biotin-Ubiquitin (0.5ug) and Smurf2 (2ug) in 50mM Tris pH 7.4 for 30min at 37C, then add to streptavidin coated plates for 30 min at 37C. Incubate with anti-Smurf2, then Europium-labeled secondary antibody, and detect by time resolved fluorescence (lambda-ex=330nm, lambda-em=620nm). Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors and selectivity profiling. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing. Store at -70C. Aliquots are stable for 6 months at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
