This antibody has been selected for its ability to neutralize human cell surface uPAR-mediated bioactivity. The exact concentration of antibody required to neutralize cell surface uPAR mediated bioactivity is dependent on the uPA concentration as well as on the number of receptors present on the cell surface (a function of cell type and assay conditions). To provide a guideline, we have determined the neutralization dose for this antibody under a specific set of conditions. The Neutralization Dose50 (ND50) for this antibody is defined as the concentration of antibody required to yield one-half maximal inhibition of the cell surface uPAR mediated uPA response on a responsive cell line. ND50: ~0.2-0.6ug/ml on U937 cells. Applications: Suitable for use in Direct ELISA, Western Blot, Immunoprecipitation and Neutralization of Bioactivity. Other applications not tested. Recommended Dilution: ELISA: 0.5-1ug/ml Western Blot: 0.1-0.2ug/ml The detection limit for rhuPAR is approximately 20ng/lane under non-reducing and reducing conditions. IImmunoprecipitation: 1ug/10e6 U937 cells Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
