VEGF (vascular endothelial growth factor) is a homodimeric, disulfide-linked glycoprotein involved in angiogenesis which promotes tumor progression and metastasis. It exhibits potent mitogenic and permeability inducing properties specific for the vascular endothelium. Multiple isoforms of VEGF containing 206-, 189-, 165-, and 121-amino acid residues have been described which arise from alternative splicing of a single gene. The smaller two isoforms, VEGF165 and VEGF121, are secreted proteins and act as diffusible agents, whereas the larger two remain cell associated. VEGF/VPF plays an important role in angiogenesis, which promotes tumor progression and metastasis. Applications: Suitable for use in Neutralization, Immunohistochemistry, ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA (Sandwich): 0.5-2ug/ml (100ul/well). Used in conjunction with V2110-03 Vascular Endothelial Growth Factor (VEGF) (Biotin) allows detection of 0.2-0.4ng/well of recombinant human VEGF. Western Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit is 1.5-3.0ng/lane, under either reducing or non-reducing conditions. Neutralization: 0.05-0.1ug/ml yields one-half maximal inhibition [ND50] of the biological activity (10ng/ml). Immunohistochemistry (Formalin-fixed paraffin embedded): 2.5-5.5ug/ml with a 30 minute incubation at RT or an overnight incubation at 4C. Requires HIER using sodium citrate buffer pH6.0 prior to staining Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
