VEGF (vascular endothelial growth factor) is a homodimeric, disulfide-linked glycoprotein involved in angiogenesis which promotes tumor progression and metastasis. It exhibits potent mitogenic and permeability inducing properties specific for the vascular endothelium. Multiple isoforms of VEGF containing 206-, 189-, 165-, and 121-amino acid residues have been described which arise from alternative splicing of a single gene. The smaller two isoforms, VEGF165 and VEGF121, are secreted proteins and act as diffusible agents, whereas the larger two remain cell associated. VEGF/VPF plays an important role in angiogenesis, which promotes tumor progression and metastasis. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Direct ELISA: 0.5ug/ml using 100ul/well antibody solution. Allows the detection of 0.2-0.4ng/well of recombinant hVEGF. Western Blot: 0.1-0.2ug/ml. The detection limit for recombinant hVEGF is 1.5-3.0ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
