Vimentin is the main intermediate filament protein in mesenchymal cells and thus is of value in the differential diagnosis of undifferentiated neoplasms. In tissue sections, this reagent shows positive reaction on connective tissue, endothelial cells, vascular smooth muscle cells and astrocytes. In tissue culture typical intermediate filament profiles are seen in fibroblasts and other cell types, which contain vimentin. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:500 detects Vitenmin on 10ug og HeLa lysates. HeLa lysate was resolved by electrophoresis, transferred to PVDF membrane and probed with V2122-08. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunohistochemistry: 5-10ug/ml, Can be used on formalin fixed paraffin embedded tissues with traditional high temperature, citrate buffer treatments. Prolonged formalin fixation can reduce antibody activity. Immunocytochemistry: 5-10ug/ml Optimal dilutions to be determined by the researcher. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
