Vinculin is a cytoskeletal protein associated with the cytoplasmic face of both cell-cell and cell-extracellular matrix adherens-type junctions, where it is thought to function as one of several interacting proteins involved in anchoring F-actin to the membrane. Both human and chicken embryo sequences of vinculin contain 1,066aa and, furthermore, the 2 proteins exhibit a high level of sequence identity (greater than 95%). Vinculinis mapped to 10q22.1-q23. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1-2ug/ml detects vinculin in cells and tissues Immunohistochemistry (frozen): 2-4ug/ml on formalin or acetone-fixed tissues Optimal dilutions to be determined by the researcher . Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
