MaxLight(TM) 490 is a new Blue-Green photostable dye conjugate comparable to DyLight(TM)488, Alexa Fluor(TM)488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm), Emission (515nm), Extinction Coefficient 73,000. Wnts have diverse roles in governing cell fate, proliferation, migration, porarity and death. Wnt signaling controls development and is important for embryogenesis and carcinogenesis. Wnts are transduced through at least three distinct intracellular signaling pathways including the canonical 'Wnt/b-catenin, 'Wnt/Ca2+ and 'Wnt polarity (also called the 'planar polarity pathway) pathways. Distinct sets of Wnt and Frizzled ligand-receptor pairs activate each of these pathways and lead to unique cellular responses. The Wnt/b-catenin pathway primarily regulates cell fate determination during development, whereas the main function of the Wnt polarity pathway is the regulation of cytoskeletal organization. The molecular weight of Wnt1 is 41kD. Wnt1 has a role in melanocyte expansion and differentiation during mouse development. Mouse proto-oncogene Wnt1 is activated by proviral insertion of mouse mammary tumor virus (MMTV). Transgenic expression of mouse Wnt1 using MMTV LTR enhancer causes mammary adenocarcinomas in approximately 50% of female transgenic mice by 12 months of age. Wnt1 induces mouse mammary carcinogenesis even in estrogen receptor alpha knockout mice. The interaction between b-catenin and EGFR/c-neu heterodimers in Wnt1-mediated breast tumor genesis is indispensable, thus it may be a fundamental signaling event in human metastatic progression. Wnt1 is expressed in human gastric, pancreatic and breast cancers. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested. Recommended Dilution: Western Blot: 0.5-2ug/ml Immunoprecipitation: 7ug/reaction Optimal dilutions to be determined by the researcher Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight(TM)490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
