The inhibitors of apoptosis (IAP) proteins prevent cell death both in vitro and in vivo by virtue of inhibition of distinct caspases. The X-linked Inhibitor of Apoptosis, XIAP, is a key member of the IAP family of proteins. The caspase inhibiting activity of XIAP is negatively regulated by at least two XIAP-interacting proteins, XIAP-associated factor 1 (XAF1) and Smac/DIABLO. XAF1 binds to XIAP and antagonizes caspase inhibitory activities of XIAP. Expression of XAF1 triggers a redistribution of XIAP from the cytosol to the nucleus. XAF1 is ubiquitously expressed in normal tissues, but is present at low or undetectable levels in many different cancer cell lines. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1-2ug/ml, A band of 34kD should be detected. Immunohistochemistry: 1:100 Optimal dilutions to be determined by the researcher. Recommended Positive Conrol: Human liver Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
