| Membrane-bound Aminopeptidase P2 is a hydrolase specific for N-terminal imido bonds. It is widely expressed in many adult tissues with the highest levels in the kidney. Applications: Suitable for use in Western Blot, Immunoprecipitation, Direct ELISA and Neutralization. Other applications not tested. Recommended Dilutions: Western Blot: 1-2ug/ml. Using a colorimetric detection system, the detection limit is ~10ng/lane under non-reducing and reducing conditions. Immunoprecipitation: 25ug/ml immunoprecipitates recombinant human X-prolyl aminopeptidase from conditioned cell medium. Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~1ng/well. Neutralization: Inhibits/neutralizes the enzymatic activity of recombinant human X-prolyl aminopeptidase. X1049-56A was preincubated with the enzyme at different molar ratios at RT for 30 minutes. The enzyme was then assayed with K(Abz)PP-pNA. 50% of the proteolytic activity was inhibited by X1049-56A at ~1.5ug/ml (IC50) under conditions in which the enzyme was present at 2ug/ml and the substrate concentration was 0.05mM. Considering the molecular masses of the enzyme (72kD) and the antibody (150kD), IC50 was achieved at ~1:2 molar ratio of the antibody to the enzyme. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
