Initial studies suggested that DAI may be involved in processes such host response to neoplasia, additional research has indicated that the cytosolic DAI can act like the toll-like receptor TLR9 by detecting cytosolic double-stranded (ds) DNA and trigger induction of type I interferon and other innate immune responses. It is thought that the binding of DAI to dsDNA enhances its association with innate immune response proteins such as the IRF3 transcription factor and the serine/threonine kinase TBK1 (also known as NAK). DAI is highly expressed in lymphatic tissues such as the lymph node, leukocytes, tonsil, bone marrow and spleen. Expression at a lower level is also seem in thymus, lung and liver. It has been demonstrated that multiple isoforms of DAI exist. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.5-1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
