The IL36 subfamily of cytokines is the most diverse, including three agonists(IL36alpha, IL36beta, and IL36Γ), one antagonist(IL36Ra), one cognate receptor(IL36R) and one accessory protein(IL1RAcP). The receptor activation follows a twostep mechanism in that the agonist first binds to IL36R and the resulting binary complex recruits IL1RAcP. IL36gamma that binds to and signals through the IL36R which in turn activates NFkappaB and MAPK signaling pathways in target cells.
