Mizoribine is an immunosuppressant nucleoside natural product first isolated from Eupenicillium brefeldianum.1 Inhibits inosine monophosphate dehydrogenase, suppressing T-cell proliferation via depletion of guanine ribonucleotides.2 Reduces cellular GTP levels in CEM leukemia cells (68% at 4 mM and 2 hours).3 Selectively attenuates the expression of monocyte chemoattractant protein 1.4 Mizoribine also acts at HSP60, and induces an HSP60/HSP10 complex which consequently suppresses IL-6 expression.5 It displays anti-proteinuric effects by restoring normal nephrin biogenesis in nephrotic syndrome models.6
