SLB1122168 is a potent (IC50 = 94 nM) second generation sphingosine-1-phosphate transporter Spinster Homolog 2 (Spns2) inhibitor. In vivo, it significantly reduced circulating lymphocyte counts but did not reduce plasma sphigosine-1-phosphate levels as with the first generation inhibitor SLF1081851 (Focus Cat 10-4395). Spns2 inhibitors represent potential alternative immunomodulatory agents to S1P receptor agonists as they are able in induce lymphopenia without drastic changes to systemic S1P levels or stimulating the S1P1-5 signaling pathway.
