Aldometanib inhibits (IC50 = 50 µM) aldolase by blocking its binding with fructose-1,6-bisphosphate (FBP).1 It activates lysosomal AMPK and mimics a cellular state of glucose starvation. Aldometanib alleviated fatty liver and nonalcoholic steatohepatitis in mice and extended lifespan. It mitigated inflammation and oxidative stress damage in LPS-treated lung fibroblasts and also inhibited NLRP3-mediated pyroptosis.2 Aldometanib inhibited hepatocellular carcinoma in mice in an AMPK-dependent manner.3 It rebalanced the tumor microenvironment allowing cytotoxic CD8+ T cells to infiltrate tumor cells.
