PD 0325901 is a potent MEK1 and MEK2 inhibitor, which inhibits MEK activity in mouse colon cells with an IC50 of 0.33 nM. PD 0325901 inhibits growth and proliferation of a variety of cancer cells in vitro, also inhibiting production of proangiogenic cytokines such as VEGF. In vivo, PD 032901 induces G1-phase cell cycle arrest and apoptosis in a cancer model and promotes myelin recovery in drug-induced demyelination in vivo in mice. PD 0325901 enhances generation of induced pluripotent stem cells (iPSCs) and in combination with CHIR 99021 and A 83-01 induces generation of ground state iPS cells from human and rat somatic cells.
