Glu-AMS is a competitive inhibitor of Escherichia coli glutamyl-tRNA synthetase (GluRS) with a Ki of 2.8 nM, and it shows weak binding ability to free GluRS. The binding affinity of Glu-AMS for GluRS increases in the presence of cognate tRNAGlu, while no change in affinity occurs in the presence of non-cognate tRNAPhe. Glu-AMS facilitates in-depth understanding of the evolutionary mechanism of GluRS: glutamate is activated only in the presence of tRNAGlu, thus avoiding futile ATP cleavage[1].
