G-5758 is an orally active IRE1alpha inhibitor with an IC50 of 38 nM for XBP1s. It is used in studies involving multiple myeloma models [KMS-11], demonstrating good tolerance in rats at oral doses up to 500 mg/kg. G-5758 exhibits pharmacodynamic effects comparable to those induced by IRE1 knockdown.
